From: Functional exploration of co-expression networks identifies a nexus for modulating protein and citric acid titres in Aspergillus niger submerged culture

Fig. 6

Representative images and quantitative analysis of conditional expression mutant macromorphology during submerged growth in citric acid production media. 1 × 105 spores/ml of each isolate were inoculated in organic acid production medium CitACM with 10% sucrose as carbon source. Cultures were grown at 220 RPM, 34 °C, for 96 h (see “Methods” section for full conditions). Representative images are depicted for triplicated experiments each consisting of duplicate replicates (a). Pelleted morphologies (any fungal structure > 500 µm2 area) are reported as a function of the total fungal area measured during image analysis, and are indicated as a percentage in parenthesis. Scale bar in the top left panel is 1 mm. Shake flask cultures were quantitatively analysed (b) using the MPD image analysis pipeline as described in Fig. 4b. Note that pelleted morphologies were almost entirely absent in the secG mutant TC4.4 0 µg/ml Dox during growth in organic acid production medium
